Ribonucleases (RNases) are the inherent enemy to all RNA related experiment, especially to those involving an extremely low concentration of RNA samples. Utilization of ribonuclease inhibitor in sensitive experiment is a common and effective practice to safe-guard RNA against potential RNase contamination. Ensuring the availability of protected, non-degraded RNA sample can enhance the sensitivity, the yield, and the successfulness of RNA related experiment. abm’s RNaseOFF Ribonuclease Inhibitor is a recombinant protein which specifically inhibits common ribonucleases, including RNase A, B, and C, with high affinity. Thoroughly tested in cDNA synthesis and RT-PCR, abm’s RNaseOFF Ribonuclease Inhibitor is proven to be an useful additive in PCR or RT-PCR as it does not inhibit reverse transcriptases’ (RTases) and DNA polymerases’ activity. This robust version of RNase inhibitor also has improved resistance to oxidation over the highly oxidation-sensitive human RNase inhibitor. RNaseOFF Ribonuclease Inhibitor is stable even under very low concentration of DTT (<1 mM DTT), making it the best choice for ultimate RNA protection.
One unit is defined as the amount of RNase Inhibitor required to inhibit the activity of 5 ng of RNase A by 50%. RNaseOFF should be added to the tube prior to the addition of other components (including the sample) to ensure that RNase inhibitory activity. Prevent inactivation of the RNaseOFF by avoiding temperatures greater than 50°C and high concentrations of urea or other denaturing agents.

Notes
- it is recommended to keep RNaseOFF on ice at all time while it is pending for use.
- it is recommended that all RNA-related/RNase-sensitive work should be setup under conditions where RNase contamination has been eliminated. Pipette tips and tubes should be treated with 0.1% DEPC.
